There is probably a reason why they stated that in their advertisement, and I would honor their wishes by doing it this way. Often, showing up in person can tick them off to the extent that your resume gets tossed in the trash no matter how good you come off. It's that whole ego thing they like to pull out, I think... other times they are just too busy and it is viewed as an unwelcome intrusion and disrespectful. Send the application along but follow up like you normally would or with a bit more intensity.

If it is a fax response it wont make any difference if you drop a copy of your CV in.
But if it is an e-mail response, they are usually collated and sorted via computer and forwarded onto the relevant person to screen the CV. If you give a paper copy in they tend to get "lost" because they are outside of the procedure.
